The title of Luis Daniel’s project: Early stages of the de novo formation and dynamic of peroxisomes in Saccharomyces cerevisiae
Luis Daniel will identify novel Saccharomyces cerevisiae proteins involved in the formation of peroxisomes from the endoplasmic reticulum using genetic (synthetic genetic array analysis) and biochemical (pull-down experiments) approaches. Promising proteins will be analyzed for their localization, function and physical interaction with known proteins involved in peroxisome formation from the endoplasmic reticulum. Luis Daniel will contribute to the isolation and structural analysis of Pex25 in collaboration with EMBL (Hamburg). Based on structural information obtained at EMBL, mutants will be designed and analyzed for their function in peroxisome formation from the endoplasmic reticulum using in vivo and in vitro approaches. In this context, Luis Daniel will also investigate the nature of newly described ER-derived preperoxisomal membrane remnants, with emphasis on the topogenesis of components of the peroxisomal protein import machinery and assembly of the peroxisomal importomer.
For identification of the origin of peroxisomes and analysis of the topogenesis of peroxins of the importomer, Luis Daniel will establish fluorescence microscopy markers to optimize imaging of peroxisomal proteins and peroxisome formation in a spatiotemporal manner taking advantage of the SNAP- and Halo-tag technology and mutants with “arrested” phenotypes as well as induction of “rescuing” proteins.
Part of the research will be performed at the RUG (Groningen)and EMBL (Hamburg).
